DSIP (Delta Sleep-Inducing Peptide) is a small nonapeptide with the sequence Trp-Ala-Gly-Gly-Asp-Ala-Ser-Gly-Glu, first discovered for its association with promoting deep, restorative delta-wave sleep. Found in the brain, hypothalamus, and other tissues, DSIP has been extensively studied for its influence on sleep architecture, stress adaptation, and neuroendocrine regulation. Unlike traditional sedatives, DSIP appears to modulate sleep naturally by interacting with the central nervous system’s neurotransmitter pathways, including GABA and serotonin systems, and may contribute to the synchronization of circadian rhythms through hypothalamic activity. Plasma levels of DSIP exhibit a circadian rhythm, peaking in the late afternoon and dipping in the early morning. Research indicates the peptide may modulate the hypothalamic-pituitary-adrenal (HPA) axis, influencing the release of corticotropin-releasing hormone and cortisol, positioning it as a mediator in stress adaptation. Clinical studies in chronic insomnia patients demonstrated higher sleep efficiency and shorter sleep latency with DSIP compared to placebo. Additional research suggests potential roles in neuroprotection, cognitive function through glutamate and acetylcholine neurotransmitter interactions, and oxidative balance modulation.
